There IS data on the amount of time it takes for ribavirin to get to full concentration AND there is data on the efficacy of ribavirin together with interferon.
Avatar n tn How important is it to be taking a full ribavirin dosage late in treatment? I am male, 39yrs, 140lb, genotype 1a, in week 34 of peg-intron+riba treatment. I started tx with a viral load of 3,000,000IU/m. Viral load tests were undetectable at 12 and 24 weeks (yay!). However, I have not been tolerating the ribavirin very well - anemia, fatigue, the usual... My starting riba dosage was 800mg/day, but I have been at that dosage for only a total of 8 weeks (out of 34 weeks so far).

Keep in mind that it can take 1-3 weeks for a change in ribavirin dose to be reflected in your hemoglobin. Also, and again based on your previous treatment experience, consider Procrit (epo) coincidental with the riba pre-dosing for same reasons.

If so, it appears to be a carefully guarded secret :) That said, the concept of pre-dosing ribavirin makes sense to this non-medical professional on a logical basis. Since it takes 2-4 week for ribavirin to reach maxium serum levels, then in a sense, if we don't pre-dose ribavirin, we are under-dosed for the first 2-4 weeks of treatment..

The Roche/Rebetol docs do note that it is mutagenenic and "should be considered a potential carcinogen" and recent research on its effectiveness against hcv indicates that at least one of the mechanisms is via mutagenesis ("Ribavirin's antiviral mechanism of action:lethal mutagenesis?", J. Mol. Med 80:86, 2002 on pubmed).

Avatar n tn Am at 24 weeks and just got results back...virus undetectable. I'm genotype 1, over 60, and long term. Dr. has me on peginterferon + ribavirin. At 12 weeks, just barely 2 log drop. Then started daily Vitamin D supplements (5000mg) after reading latest studies. Doc was hesitant but agreed. Now says was good idea. I hope this helps someone out their. Just hoping no relapse for next 24 weeks. Have had gradually increasing neutropenia but still over the <500 limit and still asymptomatic.
