t know where to post this question, someone knows if pde5 inhibitors and specifically tadalafil (cialis) are reversible enzyme inhibitors? and can you explain why from a chemical and pharmacological point of view? So, is the inhibition reversible or irreversible and why? I know it is not an easy question but I can't find on the internet this answer.
I have now have tried all the usual tablets (Viagra, Levitra, Tadalafil although not Spedra - yet). Tadalafil seems to work best although even that doesn't facilitate an erection sufficient for intercourse. So, looking for alternatives, I discovered alprostadil.
I tried sildenafil and tadalafil and both worked very well only a few first times - later the was a small or sometimes no response. My testosterone is high (810) and no smoking, not overweight, blood work works O.K.

I sincerely feel that you should consult a urologist for the same if the oral medications are not able to treat the ED. I hope you have tadalafil also apart from sildenafil. I was not having any urological problems till now exept ED which started since last 9-10 months. My doc prescribed me with peroxitene and tadalafil for ED and I completed the medication 5 days back. Now I need to urinate atleast 10-12 times a day even after drinking much less amount of water(only 1-1.5ltr/day). Till I feel the pressure of urinating so severely that sometimes I lost controll of holding before actually urinate. I am feeling helpless. Please help me get out of this situation.

At least once a year we lose control and have to stop because of the tolerance increase. 10 days ago we both stopped taking between 4-5 40mg OC per day. This past weekend we were discussing (not arguing) and the topic of tolerance came up. I said that these pills should never have been given for long term pain management because there is no way to help with the tolerance and the constant need to have more to achieve the same level of pain management. He says that is NOT true.
He believes that if it is truly only being used for pain and not for ANY kind of high then you dont build up a tolerance.
Ive been on pain meds for 5 or so years, and am curious if anyone else who has taken them for that long, has experienced them 'not working' anymore? we build our tolerance, and eventually its not the same. does that mean its time to stop taking them b/c they arent working like they used to? or wein down to get tolerance lower? does this get to a point where regardless of how hight the dosage, there comes a point where they stop working all together?

Tadalafil (Cialis) has had a few side effects with long-term use, but these have not been conclusively attributed to the drug.
Headaches, indigestion, back and muscle pains, etc., have been considered the minor side effects of this drug.
Longer-term use can cause vision impairment due to optic neuropathy and has also been reported to cause sudden deafness.
Further studies regarding the long-term use of this drug are underway and are yet to be conclusively reported upon.

So I did my 1 hour glucose tolerance last week and got my results today. Kinda upset...they came back abnormal. They like for the glucose to be under 140 and mine was 164. About 3 hours after drinking the glucola last week my sugar dropped to 42, luckily I'm a nurse and noticed what was going on and immediately checked my sugar and then ate. But anyway...I go back on Friday to do my 3 hour glucose and am so nervous it is going to come back positive too.
I take generic loratabs for pain because I have cronic/progressive pain issues, I do need them for pain but at this point I have developed a tolerance that is pushing me over the line from needed to abusing...I worry about damaging my liver and heart or simply becoming a true addict and hurting my family but I need pain relief.
So my question is this, if I am going to get back to being able to only use them when the pain is unbearable how long must I detox before I loose the tolerance?
I have been taking clonazepam 0.5 mg b.i.d. for about a year now. It seems that I have built a tolerance to the medication, and now I'm worried because I've heard that this dosage is the most effective for panic disorder / anxiety. I will talk to my doctor about this but wanted to know if anyone had any insight or suggestions. I've heard of the option of combining an SSRI with the clonazepam.
